Timothy has sisters Julia and Terry, and a brother, Buck. He attended East Lansing High School and graduated in 1975. He then pursued drama at East Tennessee State University and participated in productions with the Actors Theater of Louisville. By age 18, he had his first professional theatrical role in a children's adaptation of "A Midsummer Night's Dream." In 1981, Busfield moved to New York City and joined the Circle Repertory Company, appearing in Lanford Wilson's "Talley and Son."

Timothy made his film debut as a soldier in "Stripes" (1981) and soon moved to Los Angeles, where he starred on the ABC series "Reggie" (1983). He gained recognition with his role as Arnold Poindexter in "Revenge of the Nerds" (1984) and its sequel "Revenge of the Nerds II: Nerds in Paradise" (1987).
